The interleukin-1 (IL-1) family is composed of at least 6 proteins encoded by 3 genes. The IL-1A gene (the gene for the interleukin IL-1α), the IL-1B gene (the gene for the interleukin IL-1β) and the IL-1RN gene (the gene for the interleukin IL-1RA) are all located on the long arm of human chromosome 2. At the IL-1 locus several single-base polymorphisms are known. These gene variants either cause protein dimorphisms (e.g. IL-1A -889) and / or influence the level of IL-1 formation. IL-1B +3953, for instance, results in an increased formation of interleukin-1β. IL-1β is pro-inflammatory, a key regulator in the host responses to microbial infections and a major modulator of bone resorption.

Two of the polymorphisms in the IL-1 gene cluster (allele 2 of IL-1A -889 and IL-1B +3953) are correlated with severe adult periodontitis. All patients are at risk which carry at least in one of the both gene loci a copy of allel 2. We perform a test which allows determination of the presence of allel 2 in one or both loci, and basing on the results give an appraisal of the risk to develop periodontitis.
